DELIVERY OF CELL VECTOR WITH PEI/DECORIN NANOCOMPLEX REVEALS THERAPEUTIC POTENTIAL AGAINST RAT ANTI-THY1.1 GLOMERULONEPHRITIS

Abstract:


Decorin (DCN) is a leucine-rich proteoglycan secreted by glomerular mesangial cells (MC). It has been proven to be a promising anti-fibrotic agent to treat glomerulonephritis. In present study, we tried to use Polyethylenimine (PEI)/DCN nanocomplex to transfect MC in vitro and deliver the MC vector with PEI/DCN into kidney tissue of rat anti-Thy1.1 nephritis via left renal artery injection in vivo. The results showed that nanocomplex at N/P ratio 20 demonstrated the highest transfection efficiency and the lower cytotoxicity in cultured MC. Following left renal artery injection, the exogenous MC with DCN expressing was mainly located in mesangium and glomerular capillary. Overexpression of DCN in diseased glomeruli could result in the inhibition of collagen IV deposition and MC proliferation. The pathological changes of rat nephritis were alleviated following the injection of vector. These findings demonstrate that DCN gene delivered by PEI/DNA nanocomplex with MC vector is proved to be a promising therapeutic method for glomerulonephritis.
